The honest answer upfront
NMN (nicotinamide mononucleotide) is one of the most heavily marketed longevity supplements of the past decade — and one of the most overhyped. Yes, it raises blood NAD+ levels in humans (Grade B evidence). No, there is no human evidence that this translates to reversing ageing or extending lifespan (Grade D). Our verdict: skip it for now — not because it is definitively useless, but because the proof does not yet match the price.
What NMN is
NMN is a precursor to NAD+ (nicotinamide adenine dinucleotide), a coenzyme involved in cellular energy production, DNA repair, and the activity of sirtuins — proteins linked to metabolic health and ageing in animal models. NAD+ levels decline with age, and the hypothesis driving NMN's popularity is that replenishing them could slow or reverse age-related decline.
The logic is mechanistically plausible. In animal models — mostly mice — NMN supplementation has produced impressive results: improved insulin sensitivity, better endurance, reduced markers of inflammation, and extended healthspan. But mice are not people, and this is where the hype has consistently outrun the evidence.

This is the one claim NMN reliably delivers. Multiple short-term human trials have confirmed that oral NMN at doses of 250–900 mg/day significantly raises circulating NAD+ levels. A 2023 review published in Advances in Nutrition (covering all available human RCTs) confirmed consistent NAD+ elevation across different doses and populations. This is not disputed.
What remains entirely open is whether higher blood NAD+ translates into any meaningful health or performance outcome in humans over the timescales most people care about.
Physical function and endurance — Grade C
A small number of short-term human trials have reported modest improvements in aerobic capacity and physical performance. One 2021 trial in recreational runners found that NMN improved muscle oxygen utilisation compared to placebo. Another study in older adults reported improvements in gait speed and muscle strength. These are genuinely interesting signals — but every trial to date is small (typically under 50 participants), short (six to twelve weeks), and funded or conducted by groups with commercial interests in the outcome. They are hypothesis-generating, not conclusive. Grade C: limited, early-stage, requires replication.
Reversing ageing or extending lifespan in humans — Grade D
Zero human evidence. The longevity claims attached to NMN come almost entirely from animal studies, most of which used very high doses relative to body weight that do not translate directly to a human equivalent. It is not a proven risk, but it is not a dismissed one either. Anyone with a cancer history should not self-supplement without specialist advice.
On the regulatory side: in the UK and EU, NMN has not been authorised as a novel food. This means many products are technically in a legal grey zone, and the manufacturing and quality standards applied can be inconsistent. This is not a trivial concern — supplements in regulatory limbo often suffer from poor quality control.
How to buy NMN well — if you go ahead
If you decide to experiment despite the Grade D longevity evidence, apply the same discipline you would to any unregulated market:
- Demand a certificate of analysis (CoA) from an independent third-party laboratory — not a brand-owned lab.
- Look for brands that publish heavy-metal testing results.
- Avoid products that make explicit anti-ageing, disease-prevention or lifespan-extension claims — these are not supported by evidence and may indicate a willingness to overreach elsewhere.
